Validation of fetal microchimerism after pregnancy in the ovine using qPCR

چکیده

Abstract Fetal microchimerism has been detected in maternal tissues of humans and rodents during after pregnancy. Studies focusing on fetal DNA transfer to domestic animals are limited, especially sheep. ram was observed the circulation pregnancy, but it is not known if this chimerism persists soft parturition. The objectives exploratory study were to: 1) determine male detectable mature ewes parturition so, detection repeatability differed with lifetime offspring sex ratio 2) present yearling (primiparous) shortly Eight (open, non-lactating) 8 (primiparous, periparturient) Rambouillet used. Mature (5- 7-yr old) had given birth primarily 82% males (n = 4) or 71% female over a lifetime. Yearling birthed either singleton lambs. extracted from 10 11 different ewes, respectively. Real-time PCR (qPCR) used identify presence SRY gene each tissue sample. Male brain liver one open ewe that two six females her In younger gave lamb, thyroid pancreas second ewe. ovary lamb. Based these data, we suggest possible sheep may remain pregnancy ended. associated offspring. collected greater found be due having enough time for clearance occur. Future studies warranted further XY its potential role ovine physiology.

Fetal cellular microchimerism in miscarriage and pregnancy termination

Fetal cells transfer to the mother during pregnancy and can persist long-term as microchimerism. Acquisition of microchimerism may also occur during pregnancy loss, either miscarriage or pregnancy termination. Because nearly half of all pregnancies end in loss, we recently investigated the magnitude of fetal cell transfer during pregnancy loss and whether obstetric clinical factors impacted cel...

Pregnancy Prediction in Single Embryo Transfer Cycles after ICSI Using QPCR: Validation in Oocytes from the Same Cohort

Cumulus cell (CC) gene expression is being explored as an additional method to morphological scoring to choose the embryo with the highest chance to pregnancy. In 47 ICSI patients with single embryo transfer (SET), from which individual CC samples had been stored, 12 genes using QPCR were retrospectively analyzed. The CC samples were at the same occasion also used to validate a previously obtai...

Fetal microchimerism and cancer.

The persistence of fetal stem cells with multilineage potential in women who have been pregnant, a phenomenon known as fetal microchimerism, is emerging as a potential contributing factor in certain diseases, including cancer. For example, fetal microchimerism has been implicated in autoimmune disease, wound healing, and cancer. Studies of this phenomenon may provide a novel perspective on canc...

Cellular fetal microchimerism in preeclampsia.

Previous studies have shown elevated concentrations of free fetal DNA and erythroblasts in maternal circulation in women with preeclampsia compared with those with normal pregnancy. Pluripotent and immunocompetent fetal cells also transfer to the maternal circulation during pregnancy, but whether concentrations of fetal mononuclear cells also differed in preeclampsia was unknown. We sought to q...
